the other day I had some time to work on one of my tvs that I have the lumagen radiance connected too.
I was doing the autocal with the x-rite display3pro color meter.
I would like to know can I leave the wait time set at zero or do I need to bump it up to 400 or 600ms so the color meter has time to take a reading before the test patterns change the next one.
and if so how do I go about finding out how to set the delay for my meter is there a test in the software I can use,

I would leave it at zero. The delay is there only to deal with unusual situations. Ordinarily, it is not needed.
